"New" Therapy of Cachexia : Through "One" stone (15K) catching "Two" birds (PAK1 and COX-2).

Cachexia is a complex "body wasting" disease, an acute ageing process, apparently depending on three "oncogenic" enzymes (PAK1, TOR and COX-2) at least.
A decade ago, we developed a highly cell-permeable ester called "15K" from an old pain-killer called Ketorolac, a racemic mixture of R- and S- Ketorolac, through CC (Click Chemistry).
15K is 500 times more potent than Ketorolac, to block the PAK1-dependent growth of lung cancer cells, while 5, 000 times more potent to block COX-2 dependent growth of B16F10 melanoma cells. R-Ketorolac is a PAK1-blocker, while S-Ketorolac is a COX-2 inhibitor.

1,2,3-Triazolyl ester of Ketorolac (15K): A "Click Chemistry"-based highly potent PAK1-blocking cancer-killer. Eur J Med Chem. 126: 270-276.

Thus, it is most likely that 15K, apparently one stone, would catch two birds (PAK1 and COX-2), hopefully curing cachexia which depends on PAK1 and COX-2, 500-5,000 times more efficiently than Ketorolac.
